The Toxic Effect of Organic Acids on Escherichia coli W |

The toxic effect of 3-hydroxypropionic acid (3-HP) at high concentration on cell growth and cellular metabolism is a great challenge to its commercial production. This research has examined and compared the toxic effect of 3-HP on cell growth with other similar weak acids, especially lactic acid, under varying acid concentrations, temperatures and pHs by using Escherichia coli W as the test strain. Compared with lactic acid, 3-HP was observed to be approximately 10-fold more toxic. When the growth inhibition was correlated with undissociated acid concentration, the two acids presented no appreciable difference. The growth inhibition by other small organic acids such as was also well correlated with their pKa values or protonated free acid concentrations. This study suggests that the growth inhibition by small weak acids is mainly caused by the so-called proton effect rather than anion effect, i.e., the increase in intracellular proton concentration. A proper increment in medium pH is suggested to alleviate the acid toxicity by reducing free acid concentration in the culture medium. |
