Islet transplantation is a potential therapy for type 1 diabetes mellitus. However, they can rapidly rejected in the body due to early graft rejection of transplanted islet. Among them, most islet mass was mainly eliminated during early loss. Recently, it was reported that high-mobility group box 1 (HMGB1) protein played crucial roles in immune response. After islet transplantation, damaged islets secreted HMGB1 proteins, thereby inducing inflammatory responses by signal pathway through its receptors. Therefore, attenuation of HMGB1 activity can contribute to protection of islet from early graft loss. Glycyrrhizin has the effect of inhibition of HMGB1 activity by physical binding. So, we modified islet surface with glycyrrhizin to modulate HMGB1 protein. Glycol chitosan was conjugated with glycyrrhizin to introduce more abundant glycyrrhizin onto islet surface. In conclusion, glycyrrhizin-conjugated islets successfully captured HMGB1, thereby attenuating the HMGB1 protein activity. |
