제목 Engineering strategy to produce methionine effectively by using glucose and formate simultaneously in Escherichia coli
초록  Methionine, which is one of essential amino acids, can be used for many purposes including food additives, cosmetics, or pharmaceutical reagent. In Escherichia coli, methionine pathway is well known and methionine is synthesized from homocysteine and methyltetrahydrofolate. Because homocysteine comes from TCA cycle and methyltetrahydrofolate can be made from formate and tetrahydrofolate, I’m trying to use glucose and formate simultaneously to produce methionine effectively in Escherichia coli. For this purpose, I will perform deletion of gcvTHP and glyA genes and over-expression of ftfL, mtdA and fch genes that originate from Methylobacterium extorquens AM1. Through this engineering, Escherichia coli must use formate because of the deficiency of ability to produce methylenetetrahydrofolate. After that, additional engineering to increase methionine production will be performed.
