pH/temperature sensitive multiblock copolymer hydrogels from Poly(amino ester urethane) used for protein delivery |
초록 |
A novel series of novel multiblock copolymers contained of temperature-sensitive triblock poly(caprolactone-co-lactide) - poly(ethylene glycol) - poly(caprolactone-co-lactide) (PCLA-PEG-PCLA) and pH-sensitive cationic moiety poly(amino ester urethane) (PAEU) was developed for biomolecules delivery purpose. The synthesis of them was confirmed by 1H-NMR and characterized by GPC. For in vitro gelation, they showed the gel region that responsed to pH / temperature changes, covered body physiological condition and could be controlled by changing PEG molecular weight. The in vivo gelation behaviour showed the result of rapidly in situ gel forming after injecting subcutaneously into SD rat. The in vitro and in vivo stability of the gels were also checked. For biomolecules delivering and releasing, FITC-BSA was used as the model protein for in vitro releasing experiment. This novel copolymer hydrogel has shown the potential to be used as protein carrier system. |
