In cancer chemotherapy, effective strategy for local delivery and sustained release of therapeutic genes and drugs are necessary. In our study, we developed a nanofiber with gene delivery efficiency containing anti-cancer drug to treat liver cancer. Here we report that the nanofibers mainly compounded by branched poly (ε-caprolactone) (PCL) macro-monomers and anti-cancer drug paclitaxel. An administration of this nanofiber over the liver tumor site will dramatically hinder the cell growth and prevents metastasis. The size of paclitaxel encapsulated PCL (PTX/PCL) nanofibers was determined by SEM image. For gene delivery, cationic polymer was used to prepare nanoparticle with miRNA 145 and later it was coated over the nanofiber. The nanofiber showed localization and time-dependent sustained release of the microRNA for liver cancer cell uptake. From this study, we are expecting a synergistic effect on the cancer cell suppression since we have combined the drug and gene delivery. |
