Detection of specific biomolecules based on Molecular Imprinted Hydrogel structures |

Molecular Imprinting (MI) uses a functional monomer to form a specific recognition site through covalent or non-covalent bonding with a target molecule, and then removed the target molecule so that rebinding occurs at that site. Recently, many researchers are focusing on MI using macromolecules of complex structures such as proteins. In this study, we report that Horseradish peroxide (HRP) expressing fluorescence using H2O2/Amplex red is imprinted in a hydrogel composed of Acrylamide (AAm)/N-isopropylacrylamide (NIPAm). The reason for using NIPAm and AAm is to increase affinity through interaction with the target molecule. The elution process is necessary before HRP is rebound in the HRP imprinted hydrogel (h-MIH). The fluorescence intensities of HRP imprinted hydrogel (h-MIH) and HRP non-imprinted hydrogel (h-NIH) are compared to confirm the binding affinity. To confirm the selectivity and reproducibility, binding capacity of h-MIG and h-NIG are compared by quantitative analysis. |
