Alginate-coated thiolated chitosan microsphere for an oral drug delivery system |

Thiolated polymers are reported to have mucoadhesive property owing to thiol bearing side chains. Thiomers mimic the natural mechanism of secreted mucus glycoproteins, which are also covalently anchored in the mucus layer by the formation of disulfide bonds-the bridging structure most commonly encountered in biological systems. In this study, thiolated chitosan microspheres (TCMs) were prepared by ionic gelation process with tripolyphosphate and then, the bovine growth hormone (BGH) as a model drug was loaded. Finally, the BGH-loaded TCMs (bTCMs) was coated with alginate to improve the stability of drug-loaded microspheres in gastrointestinal (GI) track. The alginate-coated bTCMs were observed as spherical shapes. The average particle size of alginate-coated bTCMs was 2.69±0.39 µm and the size-distribution was shown uniformly. Also, effects of several formulation factors on drug release according to concentration of drug and alginate concentration were investigated. |
