Effect of Purified Alginate Sponge on the Regeneration of Nucleus Pulposus Cells |

Alginate is a natural polymer. In order to overcome the immune rejection of alginate, we have developed using modifed Korbutt’s method for alginate purification. In this study, purified alginate sponge were used in intervertebral disc regeneration. NP cells were seeded in purified and non-purified alginate sponge, and then cell viability analyzed by MTT assay. RT-PCR was assessed to measure mRNA expression for specific gene of NP cells in purified and non-purified alginate sponges. The alginate sponge evaluated by histology through H&E, Safranin-O and MTS staining in vivo. The MTT result of purified alginate sponges showed that the cellular viability was excellent. The RT-PCR and histology staining result, characteristic of NP cell in the purified alginate sponge better than non-purified alginate sponge. This result indicates that purified alginate sponge is useful for intervertebral disc regeneration. This research was supported by WCU (R31-20029) and MBC (0405-BO01-0204-0006). |
