O2-Nanobubble of Iron-Porphyrin conjugated Polyaspartamide for Molecular Ultrasound contrast effect |

A number of nanoscale self-assemblies have been extensively used in various biomedical and industrial application such as targeted delivery carrier for diagnosis and therapy. In this study, we prepared the oxygen-nanobubble containing ferrous porphyrins to mimic the ability of porphyrin of hemoglobin in red blood cell to gather oxygen. First, we synthesized poly(2-hydroxyethyl aspartamide)(PHEA) grated with ferrous porphyrins, which could form self-assemblies in an aqueous solution. Then, the nanobubble was formed as liposomal bubble using 1,2-Dipalmitoyl-sn-glycero-3-phosphocholine and the ferrous porphyrin-conjugated PHEA was self-assembled to induce the oxygen inside bubble. The nanobubbles were maintained less than 1 μm of diameter and stable spherical form by modulating degree of substitution of porphyrin as 5mol%. Further, the nanobubble containing the ferrous porpytin and oxygene showed enhanced photoacoustic effect as ultrasound contrast agent via the Doppler ultrasound test. |
