제목 Highly Sensitive Detection of EGFR point mutation using Dumbbell-shaped DNA probe (DDP)
초록 Analysis of circulating tumor DNA (ctDNA) provides a minimally invasive and feasible approach for the detection of epidermal growth factor receptor (EGFR) mutations for non-small cell lung cancer (NSCLC). Molecular inversion probe (MIP) is a highly specific detection platform for the diagnosis of the EGFR exon gene 21-point mutation (L858R) in ctDNA. In this study, a dumbbell-shaped DNA probe (DDP) was designed to replace a conventional MIP to separate the binding site of the target and primer. This allowed more selective and successful binding of target to the DNA probe. DDP with selective single nucleotide gap filling approach allowed only specific ligation of DNA probe upon the binding of mutated target. After the ligation of DNA probe, it was amplified by rolling circle amplification (RCA). The amplified product was detected by Real-time (RT) PCR within 1 hour. This simple and selective method has great potential for efficient single nucleotide polymorphism (SNP) detection.
