Due to the abnormal microenvironment in the wound site, some adverse conditions downregulate the therapeutic effect to inject stem cells into skin wound. In the environment of wound site, the loss of cell function caused by enzymatic trypsin treatment can lead to a decrease efficacy of transplanted stem cells. In this study, poly (β- amino ester) (PAE) known as pH-responsive polymer was newly synthesized to provide cell culture conditions suitable for stem cell. Human adipose-derived stem cells (hADSC) cultured on PAE-based culture system had similar cell adhesion and isolation compared to hADSC cultured on conventional culture system. Also, hADSC collected with saline (pH 6.0) showed similar therapeutic angiogenesis and wound regeneration compared to hADSC separated by trypsin. In summary, for future tissue engineering, pH-reactive polymer could be applied as an alternative cell culture system. |
