제목 pH-Induced Nanoparticle Formation and Release of Ribozymes with PEG-b-Poly(L-histidine)
초록 Ribozymes are catalytically competent RNAs that can be used to reduce expression of a pathogenic gene by binding and cleaving specific regions of target RNA. The gene-targeted therapy by ribozymes has a great specificity for the targeted cell and may reduce the problem resulting from systemic toxicity. After transcription of a gene in the nucleus, the mRNA transcript is transported to the ribosome, which translates the mRNA into proteins. Ribozymes inhibit gene expression by blocking the translation of mRNA into proteins. In this study, we designed polymeric delivery vehicles for ribozymes based on poly(ethylene glycol) (PEG) and poly(L-histidine). The block copolymer, PEG-b-Poly(L-histidine) was formed complexes with ribozymes in a pH-dependent manner. The release of ribozymes from complexes was monitored at pH 7.4 using ribozyme quantitation assay. Nanoparticles formed from PEG-b-Poly(L-histidine) and ribozyme at acidic pH have about 128nm in an effective diameter. The complexes released the ribozymes with a burst phase over about 10h followed by a slower, sustained release over 70h.
