Characterization of Biocompatibility Alginate-SIS Sponge |

Small intestinal submucosa(SIS) is the natural biomaterials mainly composed of collagen, glycosaminoglycan and growth factors. Tissue engineering techniques have been investigated the possibility of the application of SIS for a wound dressing. SIS solutions of 1 and 2 wt% with 0.1% pepsin in 3% acetic acid were reacted for 48 hrs and freeze-dried. They were treated with different concentrations (50, 100 mM and 200 mM) of EDC for 24 hrs and lyophilzed. Then 1 wt% alginate solution was pass through the SIS sponge and lyophilzed. Calcium chloride was used as crosslinking agent. The prepared sponges were characterized by scanning electron microscopy. The biocompatibility of NIH/3T3 with these SIS sponges was found to be acceptable at a cell. We investigated biodegradation of SIS sponge, MTT assay. From the MTT assay results, the biocompatibility of the crosslinked SIS sponge with NIH/3T3 was confirmed. |
