De-cellularized Arterial Blood Vessels as Biocompatible scaffolds for tissue engineering applications |

This study that aims to investigate the potential of decellularized blood vessels for application in tissue engineering. Blood vessel grafts are customarily fabricated through either film formation or electrospinning of polymer based materials which may convey non-full integration. Samples of rat inner carotid artery were extracted, cleaned of unnecessary connective tissues and coagulated blood and de-cellularized by submerging in 1% sodium dodecyl sulphate (SDS) at 37°C for 1 to 3 days. The decellularized constructs where then washed with distilled water to remove excess SDS. Microstructure of the natural and decellularized and recellularized constructs were examined through SEM microscopy and histological sections stained with hematoxylin and eosin and masson’s trichrome. Tensile strength of decellularized and recellularized the materials were also observed and compared with the unmodified arterial tissue sample. In-vitro biocompatibility was determined through use of bone marrow stem cells and observing 1 week cell proliferation and cell attachment behaviour through confocal microscopy of fluorescent stained samples. |
