Intervertebral discs (IVD) are specialized structures to anchor adjacent vertebral bodies conferring flexibility. There is no optimal treatment for persistent pain associated with IVD degeneration, focus has shifted toward replacement using tissue engineered IVD. Mesenchymal stem cells (MSCs) are multi-potent stem cells that differentiate into other cells. Porcine small intestinal submucosa (SIS) generates a minimal inflammatory response following implantation and has been approved for many types of biomedical applications. We developed synthetic/natural hybrid scaffolds with PLGA and SIS for giving similar atmosphere of IVD. We evaluated the scaffold with SEM, GAG and MTT which was assessed to measure cell viability from the scaffolds. In addition, we performed the histological assay from in vivo. The results showed cell viability was well grown and specific properties were strongly expressed. These results suggest the SIS/PLGA scaffold is feasible application for IVD regeneration. |
