Epidermal growth factor receptor (EGFR) 19 deletion mutation gene is often over-expressed in carcinoma patient. EGFR 19 deletion mutation is known as typical biomarker of non-small cell lung cancer, which one section in exon 19 of EGFR gene is deleted. Therefore, during the past decades many efforts to detect EGFR 19 deletion mutation gene have been demonstrated for replacing conventional diagnostic method such as PCR and tissue biopsy. We developed a method of detecting EGFR 19 deletion mutation gene in Rolling Circle Amplification (RCA), which provides high sensitivity and simple procedure including isothermal amplification. RCA is suitable for detecting EGFR 19 deletion mutation gene using liquidbiopsy that is circulating through blood system in the form of proteins, genes. Limit of detection (~50 nM) and a faster detection time (~30 min) could be achieved by introducing RCA in this study. Herein, a simple and non-invasive in-vitro detection system on device is performed. |
