In this study, we developed a 3D-printed electrophoretic DNA extraction microchip using digital light processing-stereolithography (DLP-SL) for point-of-care analysis. Printing resin was prepared using PEG-DA (MW :258) (PEG-DA-258) as a monomer, 0.6% (w/w) Irgacure819 (IRG) as a photoinitiator, and a photo sensitizer, 0.6% (w/w) 2-isopropyl thioxanthone (ITX). Escherichia coli O157:H7 (E. Coli) lysate was loaded in the souce chamber while elution solution was filled in the sink reservoir. In order to extract DNAs, 2 V was applied to the source reservoir and the sink reservoir for 5 to 20 min. Using the isolated DNA in the sink reservoir, PCR and electrophoresis was accomplished to demonstrate the extraction performance. |
