The D-allulose 3-epimerase (DAE) enzymepreparations obtained from Bacteria (Microbacterium foliorum, Agrobacteriumtumefaciens, Arthrobacter globiformis) is used to produce alluloseby enzymatic conversion of fructose. Allulose, an epimer of third carbon offructose, is converted from fructose by catalysis of bacterial D-allulose3-epimerases (DAE). It has nearly zero caloric value and 70% sweetness relativeto sucrose. It is also known to inhibit the fat accumulation and regulate theblood glucose to low level after intake. In this study, we report that thestrain isolated from the edible vegetable showed excellent convertingactivity from fructose to allulose, Its DAE activitywas characterized for the optimal pH, temperature and metal ion dependenceusing cell pellet as crude enzyme. It had the maximum epimerizing activity at75℃ and broad pH range of pH6.0~9.0 and was dependenton manganese (Mn) and cobalt (Co) ions. |
