Bioconversion of propane to propanol using recombinant Escherichia coli as a biocatalyst |

Methylocella silvestris BL2 is a facultative methanotroph that has a unique ability to grow on methane and to utilize various multi-carbon compounds. M. silvestris BL2 is able to grow on propane by the presence and expression of two gene clusters including a soluble monooxygenase and a putative propane monooxygenase (PrMO) that can convert propane to propanol. In this research, the whole gene cluster of PrMO from M. silvestris BL2 including native promoter and noncoding region was amplified by PCR and successfully cloned into broad host range vector for expression in Escherichia coli. The recombinant E. coli that heterologously expressed PrMO was found to oxidize propane at both terminal and sub-terminal positions as resulting in formation of 1-propanol and 2-propanol, respectively. |
