An Engineering Approach for Sustainable Peptide Synthesis |

The synthesis of polypeptides has been a long-standing problem in the field of prebiotic chemistry and pharmaceutical industries. Direct peptide bond formation will not occur unless high temperature or activating agents are used because of the high activation energies. Here, we found a simple system composed of hydroxy acids and amino acids that is capable of forming peptide bonds under mild conditions. Hydroxy acids form metastable oligoesters in the oscillating (hot dry/cool wet) system and transform into mixed copolymers via the ester-amide exchange reaction. A mathematical model was developed to evaluate the kinetic behavior of the copolymerization. Finally, we found that the complexity of the copolymers can be further reduced by using deep eutectic solvents as a green media. Besides our work in polypeptides synthesis, this presentation will introduce other ongoing work of our group, such as developing sustainable inks for 3D printing based on deep eutectic solvents. |
