제목 Initial burst controlled drug delivery formulation of microcapsules
초록 Here we describe the preparation of BSA-FITC-loaded microcapsules mixed with various thermo-sensitive hydrogels as controlled release system. Microcapsules consisting of a BSA-FITC core and a PLGA shell were prepared by using a mono-axial nozzle ultrasonic atomizer. The thermo-sensitive hydrogel were prepared by using aqueous solutions of MPEG-PCL, Pluronic and chitosan. The hydrogel containing microcapsules was subcutaneously injected to a rat. To determine in vivo BSA-FITC release, each formulation of BSA-FITC-loaded microcapsules in the presence of various hydrogels was monitored in vivo over time by measuring plasma BSA-FITC in rats. The hydrogel containing microcapsules showed delayed maximum of BSA-FITC, and in addition exhibited a sustained-release profile in plasma for up to 7 weeks. Therefore, the formulation in this work was prepared by relatively simple process and allowed a BSA to be released at a constant rate with minimized initial burst for a long period of time.
