In vivo release of protein-loaded microcapsules using in situ-forming hydrogels for a suppression of initial burst |

In this work, we described the preparation of BSA-FITC-loaded microcapsules using in situ-forming hydrogels as controlled release system. BSA-FITC-loaded microcapsules were prepared using a mono-axial nozzle ultrasonic atomizer with a BSA-FITC core and a PLGA shell. Injectable formations were prepared by mixing BSA-FITC-loaded microcapsules and chitosan, Pluronic, or methoxy poly(ethylene glycol)-b-poly(ε-caprolactone) solution. All injectable formations were subcutaneously injected into a rat. While monitoring in vivo BSA-FITC release, we found that the initial burst release of BSA-FITC retarded by in situ-forming hydrogels. In addition, the bioavailability of BSA-FITC from each formulation depended on the viscosities of the in situ-forming hydrogels. In conclusion, the injectable formulations in this work may provide numerous benefits as suppression of the initial burst release and a significantly extended duration of release from in vivo drug depot implants. |
