Dual ligand effect of transferrin and RGD on Avidin-Biotin interaction mediated antitumor peptide delivery |
초록 |
In this study, we designed Transferrin antitumor hybrid peptide(Tf-peptide) of Biotinylation (bio-Tf-peptide) and RGD labelled Avidin(RGD-Avidin). We employed an avidin-biotin Dualtargeting approach, in which biotinylated Tf-peptide interaction with RGD-equipped avidin (RGD-avidin-bio-Tf-peptide). RGD-avidin-bio-Tf-peptide were characterized for SDS-PAGE performed on a mini-Protein II System. The mophology of the RGD-avidin-bio-Tf-peptide were observed by Transmission elctron microscopy (TEM). Finally, the cytotoxicity and cellular uptake of RGD-avidin-bio-Tf-peptide against cancer cells were evaluated for their tumor targeting effects using the MTT assay, conforcal laser scanning microscopy(CLSM), and flow cytometry. |
