제목 The biosynthetic optimization for uridine 5-diphospho galactose
초록 The biosynthesis for UDP-Gal(uridine 5-diphospho galactose) using several glycosyltransferases of Leloir pathway has been built up. The optimization of this system is useful for producing UDP-Gal economically and efficiently. The system involves seven enzymes and is composed of three parts. 5 enzymes were expressed and the cell extracts played the roles of the rest enzymes. Acetate kinase not only converses UDP to UTP but also regenerates ATP. Acetate kinase is intrinsically enough present in bacteria, so it is used as enzyme source. But the quantity of acetate kinase has correlation with the production of UDP-Gal. At least 10 times more units of acetate kinase are used than the other enzymes for the optimization of this system. Because the high UTP concentration contributes the good yield of UDP-Gal. After the reaction the results shows 95% of the UMP was converted into UDP-Gal. This optimization is more efficient than those established before.
