An enhanced permeability and retention (EPR) effect, based on a leaky vessel with larger endothelial gaps of the tumor gives a benefit on the targeted delivery of nanoparticles. However, heterogeneity and complexity of the tumor reduce the targeting efficiency, clinically. Here, we develop intracellularly nitric oxide-generating polymeric nanoparticles (NO-NPs) that delivering nitric oxide, tumor-specifically. Nitric oxide can induce vasodilation of the tumor vessel, allowing an increase of the EPR effect. Nitrated dextran, which is reactive with reductive tumor-microenvironment, is conjugated with poly(ethylene glycol) for preparation of NO-NPs. Overall, NO-NPs increased tumor blood flow and vascular permeability after intravenous injection. |
