Coffee ring assay for detection of isothermally amplified nucleic acid targets from multidrug-resistant bacteria |

Rapid and specific detection of multidrug resistant bacteria allows for effective treatment of patients and reduces healthcare-associated costs. Here, we propose a versatile method that can detect antimicrobial resistance gene using hybridization-induced suppression of coffee ring effect. We functionalized the microbeads with oligo subsequently targeting single stranded DNA generated by isothermal amplification. In the presence of mecA – the gene specifically expressed in methicillin-resistant Staphylococcus aureus (MRSA), the suspended microbeads led to the agglomeration of particles that resist the capillary flow during evaporation and the suppression of coffee ring formation in the dried drop. We could simply observe the different distribution of particles by microscopy with target specificity. We hope that our molecular sensing strategy based on coffee ring assay could offer simple and rapid detection for diagnosing infections, overcoming many limitations in the clinic. |
