Expression and characterization of carbonic anhydrase from thermophilic marine bacteria |
초록 |
Persephonella marina EX-H1 is chemolithotrophic, thermophilic hydrogen-oxidizing bacterium from a deep sea hydrothermal vent on the East Pacific Ocean and is known to fix CO2 using the A-type reductive tricarboxylic acid (TCA) cycle. Carbonic anhydrase, PMCA, from its genome is homologue of alpha CA from Neisseria gonorrhoeae and shares 50 % identity. Codon optimized sequence has been cloned in pET vectors and its expression has been optimized in E. coli BL21DE3. The purified protein has been characterized and its application in CO2 sequestration has been studied. |
