제목 Incorporation of Freeze-dried Bone Marrow-Derived Mesenchymal Stem Cells in 3-D BCP Scaffold for Tissue Regeneration Applications
초록 It has been a trend nowadays to incorporate live stem cells in scaffolds to promote faster tissue regeneration. In this study, freeze-dried mesenchymal stem cells will be loaded in a 3-D BCP scaffold. Being able to preserve longer the stem cells in the scaffold will make it better to facilitate its application in tissue engineering and medical treatment. The BCP scaffold was characterized and its mechanical properties were tested. The cells were cultivated in the scaffold and incubated for 5 days. The scaffold with stem cells were loaded with cryoprotectant then subjected to freeze-drying. After freeze-drying, the scaffolds were stored in -80ºC freezer for 7 days. The presence of MSCs in the pores was evaluated by scanning electron microscope. The cells were then rehydrated and its viability was tested via MTT assay and confocal microscopy. Fresh mesenchymal stem cells were seeded to the scaffold to further test the biocompatibility of the freeze-dried stem cell loaded scaffold. The biocompatibility of the scaffolds was assessed by cell proliferation assays using MTT and gene expression with RT-PCR. Cell adhesion and proliferation were further characterized using SEM and confocal images.
