제목 The proliferation of human dermal fibroblast is induced by cyclic mechanical strain.
초록 Cells receive signals composed of cell binding molecules, cell-cell adhesive interactions, soluble growth factors and mechanical stimuli from their microevironments and their physiological states (for example, proliferation, differentiation, dedifferentiation and senescence) are altered in response to signals. Many studies showed that the regulation of signals is important in the construction of bioartifical tissuese/organs to be composed of cells and biocompatible scaffold. We studied whether cyclic mechanical strain induced the proliferation of newbone-human dermal fibroblasts(N-HDFs) in three dimensional culture system with 1.5% acetylated chitosan scaffold(ACS). After being seeded on ACS, N-HDFs were cultured for three days. Then, the cells were cultured to 14% cyclic strain in bioreactor for 1 week. The results of MTT assay and bromodeoxyuridine(Brdu) incorporation assay suggested that the cell proliferation at 14% stretch increased two times, compared to the proliferation of cells cultured without strain. However, the expression of fibronectin and collagen type I and the activity of metalloproteinase-2(MMP-2) were not altered by cyclic strain. By exposing N-HDFs to 14% cyclic strain for 1 week, alpha-smooth muscle actin(alpha-SMA) positive cells were not detected. The results suggest that mechanical strain can increase the proliferation of N-HDFs without the expression of alpha-SMA.
