Evaluation and Validation of Negative Pressure for Whole Organ Recellularization |

Decellularized extracellular matrix (ECM) provides a natural housing for cell to attach, grow and proliferate due the intact presence of proteins, lipids and growth factors. To date there are several reported techniques of decellularization and recellularization for whole organ as well as tissues but few of them have been completely adapted for application. In order to overcome this problem and establish a recellularization technique, this study addressed to determine the effect of inducing negative pressure for whole kidney recellularization with nonspecific cell under non-sterile condition. Cannulated rabbit kidney were decellularized by perfusion with triton X-100 and SDS solution. Multi stained mammalian bone marrow stem cell was used as cell based probe to observe the progress of recellularization process utilizing a custom negative pressure setup. Multiple test conditions were performed by several parameters of the set-up in the attempt of simulations localized cell perfusion into the decellularized kidney. Result indicates that the current configuration is capable of running under different setting and microbeads are suitable substitute for cell. This research is underway to demonstrate the feasibility of localized cell perfusion of the decellularized kidney using a custom negative pressure setup. |
