The reconstruction of corneal endothelium has been of great interest due to its potential as a tissue-engineered replacement. Demineralized bone particle (DBP), small intestinal submucosa (SIS) and silk were used for tissue engineering. We fabricated PLGA film as a control group and various percentage of DBP/PLGA, SIS/PLGA, Silk/PLGA films as an experiment group. Corneal endothelial cells (CECs) were seeded on various films, and we studied the MTT. In result of MTT, we choose the suitable biocompatible films. We confirmed that the cell viability was higher on the Silk/PLGA films through MTT. Then, we performed SEM, RT-PCR, immunohistochemistry. In this study, cell proliferation was good in Silk/PLGA films. The RT-PCR results were shown that specific mRNA expression. Also, specific markers expression was good. In conclusion, this study suggests that Silk/PLGA film may serve as potential CEC cells differentiation and structural basis for tissue engineered CEC cells. |
