Fabrication of Electrospun Nano-fiber Sheet Made of Poly(L-lactide-co-ε-caprolactone) and Gelatin for Tissue Engineering Scaffolds |

Electrospinning techniques of various biopolymers have been studied by many previous researchers for promising tissue engineering scaffolds because they can mimic ECM. We introduced biodegradable elastic PLCL (50:50) and natural gelatin as electrospinning materials. There were definite relationships between fiber diameters and the mixing ratio of PLCL/gelatin at the fixed concentration, and the mechanical properties were varied according to the mixing ratio. The biocompatibility of electrospun nano-fiber was evaluated by the initial cell adhesion within 5 hr and cell proliferation up to 5 days. Fibroblasts (NIH-3T3, ATCC) were cultured on electrospun fiber sheet and cell proliferation was estimated. We obtained the specific mixing ratio of gelatin to enhance the cell adhesion and proliferation at the early stage. This PLCL/gelatin scaffolds may be useful for the tissue engineering of tendons/ligaments due to their superior mechanical properties and cell proliferation ability. |
