Metabolic Engineering aims to purposeful design or redesign biological system for the production of commercially valuable chemicals such as biofuels, platform chemicals and biologically active compounds. Rational redesign or design of the biological systems is, however, hard to be accomplished due to the inherent complexity of the naturally occurring biological systems. Practically, the combination of rational and evolutionary approaches is useful at this stage. Although many technologies have been developed for various library constructions, screening of the best strain out of large number of library still remains as the major bottleneck. As a screening method, selection is known to be the most effective way but limited by the growth-associated selective phenotype. In this study, we developed an intracellular metabolite sensor named “riboselector” to regulate metabolic distribution. This RNA device can transform non-growth-associated phenotype into growth-associated phenotype so that phenotype of our interest can be automatically selected with minimum efforts. |
