Temporally controlled and color-switchable fluorescence activation in live bacteria |

Fluorescent labeling of proteins is crucial for bioimaging to study complex molecular mechanisms in cells. The yellow fluorescence-activating and absorption-shifting tag (Y-FAST) is a 14-kd protein, which is capable of temporal control of fluorescence signal by fluorogen binding. It can induce color switching of bacteria or mammalian cells by utilizing reversible reaction. Unlike conventional fluorescence-activatable proteins, Y-FAST is very fast and completely reversible to fluorogen. Various fluorogens were synthesized which were shown to result in photo-activation of Y-FAST in different colors upon their binding to the protein. The fluorogens were shown to reversibly bind to Y-FAST, while sequential addition of various fluorogens resulted in their spectral shifts to different colors. We plan to apply this study as a reporter system for multicolor and temporally controlled imaging of live cells as well as bacteria. |
