제목 Biomimetic peptide-grafted substrate for feeder-free culture of human pluripotent stem cells
초록 Current culture methods for human pluripotent stem cell (hPSC) expansion require animal sources such as feeder cells or matrices derived from animals. Thus, it is difficult to obtain sufficient numbers of clinical grade hPSCs due to safety issues, quality control, and high expense. Here, we developed biomimetic peptide-grafted substrates for feeder-free, xeno-free culture of hPSCs. The biomimetic surface promoted specific integrin-mediated focal adhesion and cell-cell interactions, leading to enhanced adhesion, self-renewal, and pluripotency of hPSCs. Moreover, biomimetic peptides could be immobilized to support growth of hPSCs in a surface material-independent manner. The engineered substrates developed in this study provide a chemically defined, xeno-free culture system for clonal expansion and long-term maintenance of hPSCs.
