Comparative and quantitative targeted glycomics using stable isotopic labeling (i-QTaG) |

Here, as a proof-of-concept study, we developed a stable isotopic labeling-based quantitative targeted glycomics technique for the comparative and quantitative analysis of total N-glycans using matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS). The intensities of the N-glycan peaks from quantification method showed a good linearity with the amount of the bovine fetuin glycoproteins. The ratios of intensity between the isotopically 2-AA labeled N-glycans were close to the theoretical molar ratios (1:1, 1:2, 1:5). This report demonstrated that our method has wide applications in many fields of clinical and biochemical engineering through developing 96 or 384-well plate-based i-QTaG procedures, which would be included in the following study. |
