제목 A Remarkable Enhancement by 3D hierarchical Nanostructure in a Label-free SERS Sensor for the Detection of Urinary Exosomal miRNA
초록 In prostate cancer (PC), tumor-derived urinary exosomal miRNAs have become emerging biomarkers for early diagnosis and prognosis of PC,  Herein, we present a label-free surface-enhanced Raman scattering (SERS)-based sensor for the detection of urinary exosomal miRNAs from PC clinical samples. Label-free and ultrasensitive miRNA detection with single nucleotide-level specificity was achieved by strong amplification of SERS signals which are specific to each miRNA in three-dimensional hierarchical plasmonic nanoarchitecture. This plasmonic nanostructure is constructed by self-assembly between head-flocked gold nanopillar and target-complementary DNA probes-conjugated gold nanoparticles, which creates multiple SERS hotspots in the nano-gaps between nanopillars and nanoparticles. The SERS-based sensor achieved an attomolar detection limit, abroad dynamic range (10 aM to 100 nM), and multiplex detection of target urinrary exosomal miRNAs without any labeling or amplification steps.
