제목 MPEG-PLGA Diblock Copolymer as 5-FU Drug Carriers
초록 Implantable wafer is one of the candidates to achieve long term drug release. Therefore, much attention has been given to the development of sustained drug delivery system based on biodegradable poly(lactic-co-glycolic acid) (PLGA). In this study, we focused on PLGA modified by methoxy-polyethyleneglycol (MPEG) to control release pattern of 5-FU as anti-cancer drug from the implantable biodegradable wafers. MPEG-PLGA diblock copolymers were prepared by ring-opening polymerization for a mixture of glycolide (GA) and lactide (LA) monomer with different mole ratios in the presence of MPEG as an initiator. The 5-FU loaded wafers were fabricated by direct compression method after freeze milling to disperse uniformly 5-FU into various MPEG-PLGA diblock copolymers. The release of 5-FU from wafer fabricated by various diblock copolymer was examined. In conclusion, we confirmed that the release pattern of 5-FU can be controlled by hydrophilic-hydrophobic balance between MPEG and PLGA.
