제목 The Proliferation and Differentiation of Intervertebral Disc Derived from hMSCs on DBP/PLGA Scaffold
초록 Tissue engineering strategies have the great potential to improve upon current techniques for intervertebral disc repair. However, determining a suitable biomaterial scaffold for disc regeneration is difficult due to the limitation exist. We developed synthetic/natural hybrid scaffolds with PLGA and DBP (demineralized bone particle) for give similar atmosphere of intervertebral disc. The central hypothesis is the use of optimal DBP/PLGA scaffold will influence the proliferation and differentiation of discs derived from hMSCs. We evaluated the scaffold with SEM, MTT and RT-PCR which was assessed to measure mRNA expression from the scaffolds. In addition, we performed the histological assay from in vivo. The results showed cell viability was well attached and strongly expressed their specific properties. These results suggest the DBP/PLGA scaffold is feasible application for intervertebral disc regeneration. This research was supported by WCU (R31-20029) and MBC (0405-BO01-0204-0006).
