제목 Implantation of glycyrrhizin-immobilized pancreatic islets for diabetes therapy
초록 To cure diabetes disease, transplanted islets are rapidly eliminated due to host’s immune re-sponses. Among them, high-mobility group box 1 (HMGB1) protein has been demonstrated to play a crucial role in host’s immune response. So it is possible that attenuation of HMGB1 activity can strongly contribute to successful outcome of islet transplantation. A glycyrrhizin, a glycoconjugated triterpene produced by the licorice plant, can act as an inhibitor against active HMGB1 by directly binding. Thus, we modified the islet surface with Glycyrrhizin-glycol chitosan conjugates to modu-late the release of HMGB1 protein without any damage of islets. Furthermore glycyrrhizin could enhance the function of xenogenically implanted islets. Therefore, we demonstrated the feasibility of glycyrrhizin-conjugated chitosan for successful outcome of islet transplantation. Collectively, this novel surface modification would be an effective therapy for successful outcome of islet transplan-tation.
