Isobutanol is considered as one of the promising next-generation biofuels. We developed an isobutanol producing Enterobacter aerogenes through L-valine biosynthesis pathway. Firstly, we tried to increase precursor pool and reduce major by-products. Additionally, transformation of mutant strain was conducted for isobutanol synthesis pathway. However, it is well known that microorganism hardly produce isobutanol over 2% (w/v) because of the toxic effect of medium chain alcohol. Among the many approaches, pervaporation-integrated isobutanol fermentation process was efficient enough to reduce isobutanol contents in cultivation broth. A ptsG mutant was constructed for effective utilization of lignocellulosic biomass. Toxic effects of isobutanol were alleviated by in situ recovery via a pervaporation procedure. Compared to single-batch fermentation, cell growth and isobutanol titer were improved by 60% and 100%, respectively, in the pervaporation-integrated fermentation process. |
