제목 Synthesis and evaluation of cancer cell targeted PAMAM dendrimer derivatives as gene delivery carrier
초록 Targeting ability of gene delivery carrier is one of the most important factor for target selectivity. In this study, we introduced RGD (Arginine-Glycine-Aspartic acid) tripeptide sequence to the surface of PAMAM dendrimer (Generation 4th). RGD sequence can interact with αvβ3 integrin, which is abundant in surface of cancer cells. RGD sequence also kwon as conserved sequences in many proteins that organized in the extracellular matrix such as laminin. Due to the neutral charge of RGD sequence, we further introduced L-arginine residue between PAMAM unit and RGD sequence to give a complexation ability with plasmid DNA. Synthesis of amino acids to the PAMAM dendrimer was performed using HOBt/HBTU coupling method and the transfection efficiency of synthesized PAMAM G4-R-DGR was examined in HepG2 cell line.
