제목 2-D microphase properties of 1-o-acylceramide and ultra-long chain ceramides-assembled lipid membranes
초록 In this study, we investigated how 1-O-acylceramide (1-O-acylCer) and cermide EOP with ultra-long chain (C24-C32) (ULC Cer) associate with ceramide NP (Cer NP), respectively, by characterizing their 2-D lipid membrane structures. For this, a 2-D model stratum corneum composed of ceramides/free fatty acid/cholesterol was fabricated by using the Langmuir-Blodgett trough. The π-A isotherm of the model lipid membranes revealed that 1-O-acylCer disturbed the molecular association of lipid membranes, whereas ULC Cer led to formation of closely packed membranes. Moreover, 1-O-acylCer retarded phase transition compared with ULC Cer, which was confirmed by fluorescence microscopy and atomic force microspcopy. From these findings, we expect the development of new intelligent biosensors which can more preferentially capture enzymes.
