Characterization and Release behavior of BME/PLGA Film for Nerve Regeneration. |

PLGA has been widely used as biocompatible scaffolds material to regenerate nerve tissue. We fabricated BME/PLGA scaffoldsfor neurogenesis of BMSCs and to observe the release behavior of BME. BME/PLGA scaffold were manufactured as the concentrations of 200,400,600μM,1mM of BME by solvent casting/salt leaching method. Cellular viability and proliferation were assayed by MTT test. Morphology were confirmed by SEM. The effect of BME/PLGA scaffold on the neural differentiation of BMSCs were assessed RT-PCR. The release profiles of BME were assayed using UV spectrometer. This result suggest that BME of 600 μM loaded scaffold revealed low cell viability in the results of MTT. Mophology of BMSCs changed like a neural cell in BME of 200,400μM loaded PLGA scaffold. BME was sustainly released from BME/PLGA scaffolds for 30days. We conclude that neural differentiation of BMSCs in BME/PLGA scaffold could be possible for long period.
