Protein delivery is an emerging research field containing such as intracellular uptake and stability of proteins. For protein delivery, heparin has advantages as carriers due to its affinity with proteins with heparin binding domain. Therefore, the aim of our study is to make heparin-Pluronic (HP) nano-aggregates for protein delivery. The HP copolymer was synthesized by grafting monocarboxy Pluronic onto heparin using EDC/NHS as coupling agent. The stuctrual and physico-chemical properties of HP were characterized by 1H NMR, FT IR and TGA. The HP nano-aggregates were prepared by direct dissolution method. The content of bound heparin was measured to be 0.18ug/ug of total polymer using toluidine blue assay. The critical aggregate concentration of HP was determined approximately 0.08 g/L. DLS and TEM measurements presented the mean diameter around 89 nm and spherical shape, respectively. Therefore, the HP nano-aggregate is expected to be a good candidate for protein delivery carrier. |
