Insitu conjugated GRGD- oxidized alginate/gelatin/BCP hydrogel as scaffold for bone regeneration |
초록 |
A functionally modified peptide conjugated hydrogel system was fabricated with oxidized alginate/ gelatin/ BCP to improve its biocompatibility and functionality. Sodium alginate was treated by controlled oxidation to transform the cis-diol group into an aldehyde group in a controlled manner, which was then conjugated to the amine terminus of the peptide. Oxidized alginate, which had been conjugated to the Glycine-Arginine-Glycine-Aspartic (GRGD), was then combined with gelatin to form a hydrogel composite oxidized alginate/gelatin/BCP that displayed enhanced human adipose stem cells adhesion and spreading. 1H NMR and XPS confirmed that the GRGD was successfully grafted to the oxidized alginate. In vitro study demonstrated that human adipose stem cells were proliferated and differentiation well on the GRGD conjugated hydrogel composite. A three dimension culture technique was applied to show the adipose stem cells distribution within the GRGD-oxidized alginate/gelatin/BCP scaffolds after 14 days and 28 days of incubation. These hydrogels are promising biomaterials for bone regeneration applications. |
