We developed a biodegradable poly(lactic-co-glycolic acid) (PLGA) NGC with longitudinally oriented micropatterned inner surface by combining methods with capillary force lithography and rolling the substrate up. Additionally, we introduced interconnected pores on the surface of micropatterned NGC (PP-NGC) by using salt leaching methods to provide nutrients and oxygen infiltration for and survival of migrated host neural cells. We observed enhanced results about the orientation of neurite in mouse neural stem cells and migration of PC12 cells in micropatterned surfaces compare to the flat substrates. We implanted 10 mm of porous patterned NGC with immobilization of DOPA at rat sciatic nerve for 4 weeks. After 4 weeks of implantation, PP-NGC with DOPA group showed higher sciatic function index, CMAP amplitude, muscle fiber diameter and histological analyses compare to flat NGC group. |
