제목 Neuronal differentiation of BMSCs in BME/PLGA scaffold
초록 Current tissue engineering techniques require the use of a porous biodegradable/bioresorbable scaffold, which serves as three-dimensional template for initial cell attachment and subsequent tissue formation, both in vitro and in vivo. Bone marrow stromal cells (BMSCs) can be differentiated in culture into osteoblasts, chondrocytes, and even neurons. BMSCs can be differentiated to neuronal phenotype in supplement medium with β-mercaptoethanol (BME). Porous PLGA scaffolds slowly releasing BME were prepared by means of ice particle leaching methods. Characterizations of BME contained PLGA scaffold were analyzed by SEM, mercury porosimeter, release profile and so on. Cellular viability and proliferation were assayed by MTT test. Neurogenic differential cell were analyzed by immunohistochemical staining for NSE and NF. In conclusion, we observed that BMSCs were differentiated to neuronal phenotype and BME was slowly released from PLGA scaffold.
