Atopic dermatitis (AD) is one of the immunological disorders in the epithelial skin. Topical corticosteroid cream is inconvenient due to its low accessibility to the deeper dermal layers. Also, as the excessive dose can cause some side effects, it is necessary to continuously administer with proper amount. Here, we suggest an adhesive hydrogel loaded with dexamethasone (DEX), one of the corticosteroid, to treat AD. Mesoporous silica nanoparticles with extra-large mesopores (XL-MSNs) were embedded into the polyacrylamide/polydopamine (PAM/PDA) hydrogel matrix for the enhanced adhesiveness and loading the drug. After incorporation of XL-MSNs, the mechanical and adhesive properties were significantly increased. Compared to DEX@PAM/PDA hydrogels, DEX/XL-MSN@PAM/PDA hydrogels showed much slower release profile, contributing to minimize the side effects and consistent administration of the drug. Upon its application to the skin of AD mouse model, the thickened epidermal layers, the number of mast cells, and the level of immunoglobulin E were remarkably decreased. This study indicates the drug loaded adhesive hydrogels would be used for the treatment of inflammatory skin. |
