Fatty acid and its derivatives are significant ingredients in food, chemical, and cosmetic industry. Among the compounds, various kinds of hydroxy fatty acids are arising due to diverse possibility of changes in hydroxy group to other functional groups and broaden schemes to synthesize complex chemical structures. Especially, ω-hydroxypalmitic acid is used as one of components of pseudoceramide, which can be used as skin protection and recovery. In this study, ω-hydroxypalmitic acid was produced from glucose in E. coli. In order to increase the concentration of precursor, fatty acid degradation pathway was blocked and additional overproduction target was expressed. A CYP153, a cytochrome P450 monooxygenase which hydroxylates ω-position of fatty acid, was coexpressed resulted in production of ω-hydroxypalmitic acid. Further modification in expression system of P450 led to stable expression P450, which resulted in enhanced productivity of hydroxyfatty acid. |
